<i>Ex-vivo</i> confocal Raman microspectroscopy of porcine skin with 633/785-NM laser excitation and optical clearing with glycerol/water/DMSO solution

Confocal Raman microspectroscopy (CRM) with 633- and 785-nm excitation wavelengths combined optical clearing (OC) technique was used for ex-vivo study of porcine skin in the fingerprint region. The has been performed on samples by applying a mixture glycerol distilled water glycerol, chemical penetration enhancer dimethyl sulfoxide (DMSO) during 30[Formula: see text]min 60[Formula: treatment. It shown that use CRM at 633[Formula: text]nm allowed one to preserve high probing depth, signal-to-noise ratio spectral resolution simultaneously. Comparing effect different agents showed an agent containing provides higher efficiency. Also, increase treatment time allows improve efficiency both agents. As result clearing, detection amide-III region indicating well-distinguishable structural differences between type-I type-IV collagens improved.
